Germline-encoded mycolyl lipid-reactive (GEM) T cells with conserved \u03b1\u03b2 T cell receptors (TCRs) recognize CD1b presenting mycobacterial mycolates. As the molecular basis underpinning TCR recognition of CD1b remains unknown, here we determine the structure of a GEM TCR bound to CD1b presenting glucose-6-O-monomycolate (GMM). The GEM TCR docks centrally above CD1b, whereby the conserved TCR \u03b1-chain extensively contacts CD1b and GMM. Through mutagenesis and study of T cells from tuberculosis patients, we identify a consensus CD1b footprint of TCRs present among GEM T cells. Using both the TCR \u03b1- and \u03b2-chains as tweezers to surround and grip the glucose moiety of GMM, GEM TCRs create a highly specific mechanism for recognizing this mycobacterial glycolipid.","bibjson":{"author":[{"initials":"S","lastname":"Gras","name":"Gras S"},{"initials":"I","lastname":"Van Rhijn","name":"Van Rhijn I"},{"initials":"A","lastname":"Shahine","name":"Shahine A"},{"initials":"TY","lastname":"Cheng","name":"Cheng TY"},{"initials":"M","lastname":"Bhati","name":"Bhati M"},{"initials":"LL","lastname":"Tan","name":"Tan LL"},{"initials":"H","lastname":"Halim","name":"Halim H"},{"initials":"KD","lastname":"Tuttle","name":"Tuttle KD"},{"initials":"L","lastname":"Gapin","name":"Gapin L"},{"initials":"J","lastname":"Le Nours","name":"Le Nours J"},{"initials":"DB","lastname":"Moody","name":"Moody DB"},{"initials":"J","lastname":"Rossjohn","name":"Rossjohn J"}],"identifier":[{"id":"10.1038/ncomms13257","type":"doi"},{"id":"27807341","type":"pubmed"}],"issue":[null],"journal":{"iso_abbreviation":"Nat Commun","name":""},"pages":["13257"],"title":"T cell receptor recognition of CD1b presenting a mycobacterial glycolipid.","type":"article","url":"http://www.nature.com/articles/ncomms13257","volume":["7"],"year":[2016]},"in_pmc":"N","in_pmce":"Y","open_access":"Y"},"resolution":"1.65","same_as":{"pdbe":{"url":"https://www.ebi.ac.uk/pdbe/entry/pdb/5l2j"},"rcsb":{"url":"https://www.rcsb.org/structure/5l2j"}},"species":{"common_name":"Human","match_type":"histo:assign_species","scientific_name":"Homo sapiens","slug":"homo_sapiens"},"tcr":null,"title":"Non-classical MHC Class I molecule CD1b at 1.65Å resolution","unique_chain_count":2}}
